“4DWheat”:

  • Générer les génomes-références de six espèces d’Aegilops;
  • Effectuer les analyses pan-génomiques de 50 génomes d’Aegilops et de Triticum à partir de séquences de longues-portées guidées par les génomes-références;
  • Effectuer la découverte des SNPs et analyser la diversité à l’aide de séquences courtes à faible couverture;
  • Générer les données génomiques et les analyses bioinformatiques de cinq populations provenant de rétrocroisements et de 25 populations “NAM” (Nested Association Mapping) provenant de 30 blés synthétiques dans le but d’améliorer le design des croisements et la sélection, et ce, afin d’accroître la diversité génétique en provenance du génome  “D” d’Ae. tauschii.

Plateforme canadienne de pré-selection pour l’amélioration du blé:

  • Effectuer le génotypage et le phénotypage d’une collection de plusieurs espèces de blés cultivés, de progéniteurs, de blés synthétiques et d’espèces sauvages apparentées pour la résistance aux rouilles brune et jaune, à la fusariose et à l’oïdium;
  • Effectuer des études d’association à la grandeur du génome pour les caractères de résistance aux maladies et identifier des gènes et marqueurs génétiques utiles en amélioration du blé.

Épigénétique du blé:

  • Étudier les mécanismes d’ expression des gènes dans les blés synthétiques à l’aide de séquençage d’ARN et de micro-ARN.

Cadmium dans le lin:

  • Séquencer une collection de lin variant pour sa teneur en cadmium et effectuer des études d’association à la grandeur du génome afin d'identifier les gènes et les marqueurs génétiques liés à la réduction en cadmium dans la graine.

Maladies du lin:

  • Développer des marqueurs et isoler des gènes de résistance au pasmo, à l’oïdium et à la jaunisse fusarienne du lin.

    Powdery mildew (PM) is an obligate biotrophic fungus (Podosphaera lini) causing high yield loss to flax (Linum usitatissimum L.). To date, no PM resistance genes have been fully functionally characterized. Using GWAS, we identified a locus harboring three flax homologs of the Arabidopsis RPW8 genes that confer broad-spectrum resistance to powdery mildew. Here, we characterized the gene expression profile of the three RPW8 candidate genes following PM inoculation. The data showed that, similar to RPW8.2 acting through the salicylic acid (SA) signaling pathway, the RPW8 homologs Lus10000835 and Lus10000836 are highly expressed in resistant flax lines compared to the suscpetible lines. In contrast, the flax RPW8 homolog Lus10009328 was highly expressed in the susceptible lines, suggesting a different signaling pathway from the other two homologs. RPW8.1 has been shown to activate ethylene signaling through aminocyclopropane-1-carboxylate oxidase gene isoform 4 (ACO4) and, this elevated ethylene negatively regulates the expression of RPW8.1, thereby attenuating its mediated-cell death and disease resistance when no disease is present to avoid unnecessary defense responses. We conclude that PM resistance induced by RPW8 in flax may mainly be activated through SA signaling pathways, and perhaps similarly to the ethylene signaling in Arabidopsis.
