Isoflavonoids are a group of legume-specific specialized metabolites. Certain isoflavonoids, such as glyceollin and isowighteone, are phytoalexins induced by soybean (Glycine max) plants in response to stress. An R1 MYB transcription factor GmMYB176 regulates various steps in isoflavonoid biosynthesis in soybean. However, it requires interaction with the transcription factor GmbZIP5 for its function. Overexpression of a translational fusion of GmMYB176-GmbZIP5 fusion construct in soybean hairy roots resulted into changes in metabolite levels including glyceollin, isowighteone and an unidentified O-methylhydroxy isoflavone. The goal of this research is to elucidate the structure and genetic regulation of this novel O-methylhydroxy isoflavone. Soybean hairy roots overexpressing the GmMYB176-GmbZIP5 fusion construct will be created for purification of O-methylhydroxy isoflavone for NMR study and RNA sequencing analysis. This isoflavonoid may have similar properties to other known isoflavonoids, including pharmacological importance or with a role in plant stress tolerance. Isolation and identification of this compound can open doors for development of biotechnological strategies to manipulate isoflavonoid biosynthesis in soybean or in other crops.
