Mushroom samples were collected during a botanical expedition in Arctic Quebec near Umiujaq in July. DNA was extracted from the fruitbodies and 2 gene regions were sequenced. Using automated analyses using these DNA sequences 56 species were detected, of which 70% were agarics and these were in 17 genera. Several potentially newly detected species were among these. This experiment was used for northern materials to take advantage of the freshness of these remote fungal samples that were incidental to the funded botanical work.

Abstract

Twelve plots around Umiujaq were sampled for fungi during the last week of July 2016. Total genomic DNA was isolated from 83 collections and the fungal ITS 1 and 2 regions of the nuclear ribosomal DNA were amplified and sequenced. Using a similarity threshold of 97%, the 83 ITS sequences clustered into 56 operational taxonomic units (OTUs). Almost 70% of the OTUs belong to the Agaricales, representing a total of 17 genera. About 45% of these OTUs had a conspecific match with the sequence of a single species name from voucher specimen in GenBank® public database while 28% of the OTUs had either a conspecific match with environmental sequences only or did not have any conspecific match with the ITS sequences in the database (level of similarity ˂ 97%). This pool of OTUs potentially represents new fungal taxa.
